Kamui was born into the Amayaka-Clan of Ame no kuni. His father was not part of the Amayaka-Clan and he did not inherit the Kekkei-Genkai of his mother's clan. He never knew who his father was. Kamui was neglected by his mother and when she was locked up for a mistake during a mission, the Amayaka-Clan decided to put him in foster care. He was adopted by the parents of Kasumi, and although he was never accepted by his new parents, Kasumi and he developed a sibling-like bond.
Kamui was seen as a disgrace for the clan and was told to stay hidden, he was not allowed to be educated or trained as Shinobi. When his parents and Kasumi were not at home, the young boy sneaked out and roamed through the streets of Amegakure. He took delight in watching the stray dogs and soon tried to come into contact with them. Wary at first, he adapted to their pack-behaviour and became the leader of the pack.
It was then when Konan met him and asked him to become a spy for Akatsuki, in exchange for a proper training. Kamui agreed and began to spy for the newly-found rebel organization Akatsuki.
When Akatsuki began to extinguish the Amayaka-Clan, Kamui left Amegakure on Konan's order to find two Amayaka that were reported missing.
Kamui was always accompanied by his two Kuchiyose-wolves, Yin and Yang.

Kamui found the missing Amayaka in a small village. He was able to hide his origin and acted as a Amegakure-refugee to gain their trust. To his wonder, he recognized the missing Amayaka as Kyoko and Taiki, two friends of his adopted sister, and both were not interested in continuing to fight, only seeking a peaceful life.
Kamui became friends with Kyoko and Taiki, and when Haruka was born, he became friends with her as well.
When Pain noticed Kamui's affection for the family, he found his information no longer useful and decided to kill the deserted Amayaka. Kamui was not able to stop Haruka's parents from being killed and had to watch the child leave the village, but he begged Akatsuki afterwards to spare her life, even bailing for her.
Still, Kamui continued to work for Akatsuki as spy. He was always thankful to Akatsuki, since they took him out of his clan's grasp and spared him when the clan was killed.
Years later he met Haruka again and was able to tell her about the clan the way he got to know them.
During Akatsuki's attack on Konohagakure, Kamui and Yang were killed while Yin survived. The black wolf was said to still roam the forests around the ambush months later.
